چکیده
(1) Background: Understanding the function of circular RNAs (circRNAs), a class noncoding RNA, in psoriatic skin can provide important insights into complex regulation genes contributing to pathogenesis psoriasis. (2) Methods: A novel method was applied RNA-seq datasets from 93 biopsy samples comprehensively identify circRNAs all types, i.e., canonical intron-exon junctions mRNAs and interior (i-circRNAs) regions exons, introns, intergenic regions. Selected were experimentally validated by qRT-PCR Sanger sequencing. CircRNAs with abundant differential expression identified their putative as competing endogenous (ceRNAs) analyzed an integrated analysis circRNAs, microRNAs, mRNAs. (3) Results: With comprehensive search using no information splicing signals, we systematically 179 highly skin. Many these reported for first time many differentially expressed involved versus normal or uninvolved Validation based on three additional confirmed most Experimental analyses well-known circRNA CDR1as, circRNA, i-circRNA We also that may act ceRNAs regulate mRNA psoriasis-related signaling pathways (4) Conclusions: The result study suggested are skin, have distinct characteristics, contribute pathogenesis.

Circular RNAs function as competing endogenous RNAs in multiple types of cancer
Circular (circ)RNAs, naturally formed endogenous non-coding RNAs, have received extensive attention in recent years due to their special loop structures and specific function. circRNAs are formed with covalently closed continuous loops and are mainly generated by back-splicing processes or lariat introns from exons and/or introns. Usually, circRNAs are stable, abundant, and evolutionarily conse...

MicroRNAs (miRNAs) are small RNA molecules, about 22 nucleotide long, which post-transcriptionally regulate their target messenger RNAs (mRNAs). They accomplish key roles in gene regulatory networks, ranging from signaling pathways to tissue morphogenesis, and their aberrant behavior is often associated with the development of various diseases. Recently it has been shown that, in analogy with t...
متن کاملLong noncoding RNA associated-competing endogenous RNAs in gastric cancer
Some long noncoding RNAs (lncRNAs) play important roles in the regulation of gene expression by acting as competing endogenous RNAs (ceRNAs). However, the roles of lncRNA associated-ceRNAs in oncogenesis are not fully understood. Here, based on lncRNA microarray data of gastric cancer, bioinformatic algorithm miRcode and microRNA (miRNA) targets database TarBase, we first constructed an lncRNA-...
متن کاملCircular RNAs: Biogenesis, Function and Role in Human Diseases
Circular RNAs (circRNAs) are currently classed as non-coding RNA (ncRNA) that, unlike linear RNAs, form covalently closed continuous loops and act as gene regulators in mammals. They were originally thought to represent errors in splicing and considered to be of low abundance, however, there is now an increased appreciation of their important function in gene regulation. circRNAs are differenti...
متن کاملBiogenesis of Circular RNAs
Circular RNAs are generated during splicing through various mechanisms. Ashwal-Fluss et al. demonstrate that exon circularization and linear splicing compete with each other in a tissue-specific fashion, and Zhang et al. show that exon circularization depends on flanking intronic complementary sequences. Both papers show that several types of circular RNA transcripts can be produced from a sing...
